Research Area: Signal transduction, gene transcription

Background:

cAMP response element-binding protein (CREB) is a transcription factor that regulates the transcription of a wide spectrum of genes in response  to second massagers (cAMP and Ca++) and cell growth or stress signals.  The primary binding sites ofCREBon the promoter regions are cAMP response element (CRE) and calcium response element which have a common 5’-TGACGTCA-3’ motif.  CREBcan be phosphorylated and activated by cAMP-dependent protein kinase A, calcium/calmodulin-dependent kinase, and kinases in the mitogen-activated kinase (MAPK) and extracelluar signal-regulated kinase (ERK) signal cascades.

CREB plays an important role in neuroplasticity and memory formation by controlling the expression of c-fos, brain-derived neurotrophic factor, tyrosine hydrolase and many neuropeptides.

Application:

In indirect ELISA, the antibody is reactive to serine133 phosphorylated humanCREBpeptide and reactive to serine133 phosphorylated humanCREBpeptide conjugated with KLH or BSA.  The peptide and the conjugates were coated at 1μg/ml in pH 9.5 carbonate buffer.

Other applications have not been evaluated.
