Description: Mouse monoclonal antibody (mAb) against human Thr212 Thr217 double phosphorylated Tau

Purification: Protein G affinity purified

Target Protein: Human Thr212 Thr217 double phosphorylated Tau.

Immunogen: KLH conjugated human Thr212 Thr217 double phosphorylated Tau peptide:

CGTPGSRSR{pT}PSLP{pT}PPTREPKK. The peptide is corresponding to the amino acid sequence 204-225 on the phosphorylated Tau protein.

Fusion Myeloma: Sp2/0-Ag14

Specificity: The mAb reacts to human Thr212 Thr217 double phosphorylated Tau peptide CGTPGSRSR{pT}PSLP{pT}PPTREPKK.  The reactivity with the unphosphorylated peptide CGTPGSRSRTPSLPTPPTREPKK was not detected.

Research AreaAlzheimer’s disease and other neurodegenerative diseases

Background: Tau is a highly soluble microtubule associated protein found in high concentration in neuron of central nervous system.   The main function of the protein is stabilizing the microtubule in nerve fibre (axon).  There are six tau isoforms which are formed by alternate splicing.  Hyperphosphorylation of Tau protein results in aggregation and formation of   neurofibrillary tangles, which is significantly associated with cognitive impairment in Alzheimer’s disease.   Thr217 phosphorylated Tau has been shown to be more useful as a plasma biomarker than Thr181 phosphorylated Tau.  And study also suggested that phosphorylation at Thr212 and Thr217 improved the antigenicity of the peptide and that antibody targeting the double phosphorylation increased assay sensitivity.

Applications: Indirect ELISA:  The antibody reacts withphosphorylated peptide CGTPGSRSR{pT}PSLP{pT}PPTREPKK coated ELISA plate in indirect ELISA.
