Background:
c-kit, also known as CD117, is a receptor tyrosine kinase expressed at the surface of mast cells and certain types of stem cells. c-kit is activated and autophosphorylated upon binding with stem cell factor. Activated c-kit phosphorylates downstream SH2-domain-containing signalling molecules such as PLCγ, the p85 subunit of PI3 kinase, SHP2, and CrkL. The binding is impaired when c-kit is mutated at tyrosine719. c-kit with phosphorylated tyrosine 719 binds to the p85 subunit of PI3 kinase in vitro and in vivo. C-kit is encoded by KIT gene and the gene is frequently over-expressed in gastrointestinal stromal tumor, myelogenous leukemia, and testicular germ cell tumor. Mutational loci of the gene are implicated with responsiveness to cancer drug imatinib.
Application:
ELISA: The antibody (approx. 1:64,000 diluted) can detect the above Tyr719 phosphorylated c-kit peptide coated on ELISA plate.
Other applications have not been evaluated.
